Full-length transcriptome sequencing and bioinformatics analysis of Paris

Abstract:The purpose of this study is to lay a foundation for the systematic exploration of saponin synthesis genesin Paris byobtaining the full-length transcriptome data of Paris. The third-generation sequencing technologyon PacBio Sequel platform was used.In this study,the rhizome mixed samples of Paris polyphylla Smithvar.yunnanensis(Franch.)Hand.-Mazz.,Paris fargesiiFranch.andParisforrestii(Takht.)H.Liwith significantdifferences in saponin content were deeply sequenced,and then the obtained data were comprehensivelyanalyzedbybioinformatics.A total of42771 transcriptswereobtained,of which40945( 95.73% )were successfully matched to seven functional databases,namely KEGG,NR,Swiss-Prot,TrEMBL,KOG,GOand Pfam.Through enrichmentanalysisofKEGG pathway,48genesrelatedtocholesterol synthesis were successullyidentified,andtwokindsofkey enzyme genes involved insaponin modification were specifically screened:12O cytochrome P45O enzyme genes and 53 UDP-glucosyltransferasegenes.Additionaly,2327transcriptionfactorsweredetected,belonging to71transcriptionfactor families.Atotalof27451microsatelitesequences(SSR)werefoundbyMISAanalysis,therewerethemostcoincident types with overlapping SSR,accounting for 32.88% of all repeat types.With the help of high-throughput full-length transcriptome sequencing technology,thetranscriptomeof rhizome tisueof Pariswassystematicallyanalyzed,and thecandidategens andregulatoryelements closelyrelated tosaponinsynthesis weresuccessfully excavated.Thesefindings laythe groundwork for further study onthe molecular mechanism of saponin biosynthesis in Paris.
