κB</sub> )信號(hào)通路探討明君散對(duì)動(dòng)脈粥樣硬化(AS)大鼠的影響。方法:采用高脂飼料聯(lián)合維生素 <img src="/qkimages/zxxn/zxxn202514/zxxn20251408-1-l.jpg" with="17px" style="vertical-align: middle;"> 法建立大鼠AS模型,分為空白組、模型組、西藥組[阿托伐他汀 1mg/(kg?d) ]、明君散低劑量組 2.35mg/(κg?d) ]、明君散中劑量組 4.70mg/(kg?d) ]、明君散高劑量組 [9.40mg/(kg?d)] ,每組10只。灌胃給藥8周后,檢測(cè)血清總膽固醇(TC)、三酰甘油(TG)、高密度脂蛋白膽固醇(HDL-C)、低密度脂蛋白膽固醇(LDL-C)水平;采用蘇木精-伊紅(HE)染色觀察大鼠胸主動(dòng)脈組織形態(tài)學(xué)變化;實(shí)時(shí)熒光定量聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)檢測(cè)大鼠主動(dòng)脈TLR4、MyD88、NF- ?κB mRNA表達(dá)水平;蛋白免疫印跡法(Western Blot)檢測(cè)大鼠主動(dòng)脈TLR4、MyD88、NF- Π<sub>κB</sub> 蛋白表達(dá)水平。結(jié)果:與模型組比較,西藥組和明君散中劑量組、明君散高劑量組血清TC、TG,LDL-C水平降低,HDL-C水平升高( P<0.05 或 P<0.01 );西藥組和明君散各劑量組大鼠主動(dòng)脈組織中TLR4、MyD88、NF- κB mRNA和蛋白表達(dá)水平均降低( P<0.05 或 P<0.01 )。結(jié)論:明君散可能通過抑制TLR4/MyD88/NF- ?κB 信號(hào)通路發(fā)揮抗動(dòng)脈粥樣硬化作用。-龍?jiān)雌诳W(wǎng)" />

AbstractObjective:ToxploretheefectofMingjunPowderonatheroscleroticratsbasedonteTollikereceptor4(R4)eloid differentiation protein 88(MyD88)/nuclear factor- κB(NF-κB) signaling pathway.Methods:The atherosclerotic model of rats was established byhigh-fat feed combinedwith vitamin method.The rats were divided into the blank group,the model group,the Western medicine group[atorvastatin 1mg/(kg?d)] ,the low-dose Mingjun Powder group[2.35 mg/(kg?d)] ,the medium-dose Mingjun Powder group[4. 70mg/(kg?d)] and the high-dose Mingjun Powder group [9.40mg/(kg?d)] ,with 10 rats in each group.After gavaging for eight weeks,the levels of serum totalcolestes)otit detected.Hematoxylineosin(HE)stainingwasusedtoobservethehistologicalmorphologicalchangsofthethoracicaortainratsTe expressions of TLR4,MyD88 and NF- κB mRNA in the aorta of rats were detected by reverse transcription polymerase chain reaction (RT-PCR).The expressionsof TLR4,MyD88,and NF-κB proteinsintheaorta of ratsweredetected byWestern Blot.Results:Compared withthe modelgroup,thelevelsofserumTC,TGandLDL-Cin theWestemmedicinegroup,themedium-dseMingjunPowdergroup, and the high-dose Mingjun Powder group decreased,and the level of HDL-C increased P<0.05 or P<0.01 ).The expressions of TLR4, MyD88, NF- κB mRNA and protein in the aortic tisues ofrats in the Westem medicine groupand each dose Mingjun Powder groupsall decreased( P<0.05 or P<0.01 ).Conclusion:Mingjun Powder might playananti-atherosclerotic efect by inhibiting the TLR4/MyD88/NF-kB signaling pathway.
