Abstract:Honey is rich in nutrientsand widely cherished.As the world's largest honey producerand consumer, China faces a growing issue with adulterated honey entering the market due to increased demand. Existing nucleic acid detection systems for honey plants often fail to distinguish related species due to the lack of target specificity. This study employed bioinformatics methods to addressthis isue by comparing the highthroughput sequencing data of Robinia pseudoacacia with the genomes of related species and the NCBI NT nucleic acid database. We successfully identified a 3141 bp genome-specific DNA fragment of R. pseudoacacia. Based on this DNA fragment, we designed a PCR primer pair, RpsF1/RpsR13,with an amplification length of 257 bp. We then established a PCR system for the qualitative detection of locust honey. This system effectively distinguishes R. pseudoacacia from related Leguminosae species,such as Erythrina variegata. The system's sensitivity is notable, detecting a minimum template amount of 8.55 ag in a 20μL (2號 reaction, equivalent to 2.0 DNA copies. Testing of 20 locust honey samples yielded a 100% positive detection rate,demonstrating the system's high specificity and sensitivity.This new PCR detection assay ofers a robust technical solution for identifying authentic honey,thus helping to address the issue of adulterated honey in the market.
