Effects of supplementary clones on genetic diversity of Pinus elliottii seed orchard

Abstract:Supplementingclonesinseedorchardswith limited or mising clonesrepresents a crucial scientific management strategy.This research focusedonthe clonal seedorchardof Pinuseliotiiat Nanning Forestry Research Institute,examining howadditional clones influence theorchard's genetic diversitytoofer insights foritsscientific management.Utilizing 16 SSR markerpairs,thestudyassessed genetic diversityvariationspre-and post-clone augmentation,analyzing material geneticuniquenessand relationships through fingerprintand genetic clustering.The results were as follows:(1)Enhancing clone numbers from 18 to 5O led to a 2 . 3 7 % rise in polymorphic information content,a 1 4 . 0 7 % rise in average number of observed alleles,a 1 . 2 3 % risein average number of effective alleles,and a (204號(hào) 3 . 5 1 % rise inthe Shannon index.(2)It was determined that11 SSR marker pairs could identify all5O clones,for which fingerprnt maps of 5O clones were established.(3)The genetic distance among these clones varied between 0.018 and 0.670,and using a O.251 threshold,the UPGMA clustering diagram grouped them into seven categories. In conclusion, it is considered that clone supplementing can enhance the genetic diversity of P .elliottii seed orchards,but the extent of improvement is limited.Developing fingerprints ofers a dependable method for identifying premium P elliottii clonesand analyzing seed orchard progeny parentage.The genetic clustering of established clones serves asa referene for parent selection in P .eliottii hybridization experiments,efficiently preventing inbreeding.
